چکیده

THE DETERMINATION of glucose by the glucose oxidase-peroxidase system has been simplified by Hill and Kessler (2) with the use of an automated system first described by Skeggs (1). Wincy and Marks (3) have also developed a method using o-tolidine in the automated system. These procedures require a large quantity of sample and use of a dialyzer to separate the protein before color development. The direct micro glucose oxidase manual method developed by Kingsley and Getchell (4) for the determination of glucose in plasma without protein removal or dialysis has been adapted for use with the present AutoAnalyzer equipment.* In all of the methods abo e, glucose oxidase promotes the oxidation of glucose by molecular ox gen to gluconic acid and hydrogen peroxide. The hydrogen peroxide formed is used to oxidize o-dianisidine to an amber color in the presence of peroxidase (Fig. 1). The o-dianisidine dye has been combined with the enzyme mixture to give only one reagent to mix with the plasma as it passes through the automated system. Several drops of Tween 20 are added to the reaction mixture

A Simple Image Analysis Method for Determination of Glucose by using Glucose Oxidase CdTe/TGA Quantum Dots

Glucose, as the major energy source in cellular metabolism, plays an important role in the natural growth of cells. Herein, a simple, rapid and low-cost method for the glucose determination by utilizing glucose oxidase and CdTe/thioglycolic acid (TGA) quantum dots (QDs) on a thin layer chromatography (TLC) plate has been described. The detection was based on the combination of the glucose enzym...

Determination of blood glucose using an oxidase-peroxidase system with a non-carcinogenic chromogen.

Manual and automated methods for the determination of blood glucose have been devised using an oxidase/peroxidase system, with dl adrenaline, a non-carcinogen, as oxygen acceptor. The manual technique employs a stable single solution protein precipitant and the other reagents used are also stable. The automated methods are operated at 40/hr sample speed and washover between samples, over a very...

Determination of glucose in biologic fluids with an automated enzymatic procedure.

An automated procedure for the enzymatic determination of “true” glucose with the glucose oxidase-peroxidase system is described. A threefold increase in sensitivity was obtained through meaurement of the stable, pink chromogen obtained at high acid concentration. Good reproducibility and stoichiometry was obtained over the range of 50-400 mg./l00 ml. at a sampling rate of 40/hr. The normal ran...

Evaluation of an automated glucose-oxidase procedure.

We evaluated a slight modification of the automated oxidase/peroxidase method for glucose determination in blood with use of 4-aminophenazone [J. Clin. Pathol. 22, 246 (1969)]. Nonsugar reducing substances did not interfere, except for ascorbic acid in very high concentrations. Lipemic sera gave slightly lower values. Values for reproducibility, specificity, and sensitivity were closer to such ...
